研究莱菔硫烷(sulforaphane,SFN)对ERK、p-ERK蛋白表达的影响,探讨其诱导人肝癌HepG-2细胞凋亡的机制.采用Western Blot方法检测人肝癌HepG-2细胞内ERK及p-ERK蛋白的表达.结果表明,10、20、40μmol/L的SFN作用人肝癌HepG-2细胞48h可显著下调HepG-2细胞内p-ERK蛋白的表达,而对ERK的表达无明显影响.提示SFN可抑制ERK信号通路,这可能是其诱导HepG-2细胞的凋亡的机制之一.
To study the effect of sulforaphane (SFN) on the protein expression of ERK and p-ERK in human hapetocelluar carcinoma HepG-2 cells and to explore the inducing apoptotic mechanism of SFN on HepG-2 cells. Western Blot assay was employed to detect the expression of ERK and p-ERK proteins. The results showed that after treated by SFN at the dosage of 10, 20, 40 μg/mL for 48 h, the expression of p-ERK proteins was evidently down regulated, meanwhile, there was no remarkable change on the expression of ERK protein. A conclusion can be drawn that SFN can inhibit ERK pathway of HepG-2 cells, which may be one of the mechanism of SFN induced apoptosis of HepG-2 cells.
