本试验旨在筛选并分析绵羊肝脏组织内与α-TTP相互作用的蛋白。选用健康无病、生长发育良好的滩羊公羊,取其肝脏组织.首先通过免疫共沉淀技术分离绵羊肝脏中α-TTP蛋白复合体,然后利用质谱分析技术来鉴定蛋白复合体的成分,并通过Mascot蛋白质谱检索软件来分析筛选到的α-TTP的互作蛋白。结果表明:利用免疫共沉淀联合质谱分析技术共筛选到10个与α-TTP相互作用的候选蛋白.通过对蛋白功能的进一步分析后发现α-TTP候选互作蛋白主要为同转运、能量代谢、基因表达调控和应激相关的蛋白。由此推测.在绵羊肝脏组织中α-TTP与多种蛋白质有相互作用.协同参与维生素E的转运调控。
The experiment was conducted to screen and identify theα-TTP-interacting proteins in ovine hepatic tissue. The liver was collected from local male Tan sheep and the α-TTP complex was first purified from ovine liver via the method of co-immunoprecipitation, then the components of the complex were identified by mass spectrometry and the function of each protein was analyzed using Mascot sofeware. 10 proteins were shown to interact with ovine α-TTP and these proteins were mainly related to transportation, energy metabolism, gene expression regulation and stress. We speculate that the ovine α-TTP coordinate with a variety of proteins in liver to regulate the transportation of vitamin E.
