目的:研究蜂毒素对人脐静脉内皮细胞ECV304生长的抑制作用及其机制。方法:将人脐静脉内皮细胞ECV304进行体外培养。四甲基偶氮唑盐(MTT)法检测蜂毒素对ECV304细胞的增殖抑制作用;流式细胞术测定细胞周期;膜联蛋白Ⅴ(AnnexinV)-异硫氰酸荧光素(FITC)+碘化丙啶(PI)双参数检测细胞凋亡;免疫细胞化学法检测经不同浓度蜂毒素作用后ECV304细胞VEGF和bFGF的表达。结果:蜂毒素可显著抑制人脐静脉内皮细胞的增殖生长,并且呈浓度依赖性,其24h、48h和72h的IC50分别为5.11μg/ml、4.68μg/ml和4.40μg/ml。经蜂毒素作用后,S期细胞比例明显增加,G0/G1期细胞比例减小,细胞周期阻滞于S期,并可诱导细胞凋亡。蜂毒素可明显下调人脐静脉内皮细胞VEGF和bFGF的表达(P〈0.05和P〈0.01)。结论:蜂毒素在体外能够抑制人脐静脉内皮细胞增殖和合成VEGF及bFGF的功能,其机制可能与阻滞细胞周期、诱导细胞凋亡有关。
Objective:To investigate the inhibitory effect of Melittin on growth of human umbilical vein endothelial cells ECV304, and its mechanisms. Methods:The human umbilical vein endothelial cells ECV304 were cultured in vitro. The inhibition rate of growth of cells treated with Melittin was evaluated by MTT assay. Apoptosis was detected with AnnexinV - FITC + PI dual parameter. Cell cycle was analyzed by flow cytometry. The Expression of VEGF and bFGF in ECV304 cells was detected by immunocytochemical method. Results: Melittin had obvious inhibitory effect on the growth of human umbilical vein endothelial cells in a concentration - dependent manner, it's 50% inhibitory concertration (IC50) of 24 h, 48 h and 72 h were 5.11μg/ml, 4.68 μg/ml and 4.40 μg/ml respectively. Compared to the control group, the percentage of the cells of treatment group in S phase increased, on the contrary the percentage of the cells in G0/G1 phase decreased, and the cells were arrested at the S phase of the cell cycle with the increased rate of cell apoptosis. The protein expression of VEGF and bFGF in ECV304 cells of treatment group was significantly lower than those of the control group ( P 〈 0. 05 or P 〈 0.01 ). Conclusion: Melittin could inhibite the proliferation of human umbilical vein endothelial cells in vitro, its mechanisms were probably associated with cell cycle arrest and cell apoptosis. Melittin could downregulate the expression of VEGF and bFGF in ECV304 cells.
