目的:探讨晚期糖基化终产物受体(RAGE)、核转录因子-κB(NF-κB)基因表达与非缺血性糖尿病足发病的关系。方法:获取非缺血性糖尿病足患者及外伤截肢对照患者的足部全层皮肤组织各一块,采用实时PCR和Western蛋白质印迹的方法检测组织中RAGE、NF-κB的基因表达。结果:糖尿病足标本中可检测到RAGE和NF-κB的mRNA和蛋白质表达,其表达水平与对照组间差异有极显著统计学意义(P〈0.01)。结论:RAGE、NF-κB介导的氧化应激及炎症反应是非缺血性糖尿病足发病中的重要病理过程。
Objective : To study the relationship between the gene expression of advanced glycation end products ( RAGE), nu- clear factor -KB(NF-KB) and the pathogenesis of non-ischemic diabetic foot. Methods :The full-thickness skin tissue on the foot of non-ischemic diabetic foot patients was obtained and patients were controlled with traumatic amputation, using real-time poly- merasechain reaction( Real - Time PCR) and Western blotting for the detection of tissue RAGE, NF-KB gene expression. Results : The mRNA and protein expression of RAGE and NF-KB could be detected in the diabetic foot specimens,and the difference was obviously significant compared with control group(P 〈 0. 01 ). Conclusion:RAGE and NF-KB gene - mediated oxidative stress and inflammatory response in tissue were important pathology courses on the pathogenesis of non - ischemic diabetic foot.
