目的 观察四嗪二甲酰胺(ZGDHu-1)体外抑制肺癌细胞株A549、EBC-1、MA增殖并诱导细胞凋亡作用.方法 将不同浓度的ZGDHu-1与A549、EBC-1、MA细胞在体外培养,用锥虫蓝染色、噻唑蓝(MTT)比色法、5'-溴-2脱氧尿苷(HrdU)-酶联免疫吸附试验(ELISA)法观察ZGDHu-1对A549、EBC-1、MA细胞增殖的抑制作用 用细胞形态学、DNA含量及细胞周期分析、Annexin-V/PI双标记、Hoechst33258荧光染色观察细胞凋亡.结果 ZGDHu-1能抑制A549、EBC-1、MA细胞增殖和活力,呈现作用时间和剂量的量效关系.A549、EBC-1、MA细胞经ZGDHu-1作用48 h后,其IC50分别为165、106、210μg/L 72h后的IC50分别为42、18、90μg/L.大部分细胞阻滞于G2~M期 出现典型的细胞形态改变,亚G1峰检出并显著增加,Annexin-V+/PI-表达升高,细胞内DNA片段含量增加,Hoechst33258荧光染色后出现凋亡细胞的特征性改变,证实ZGDHu-1能诱导A549、EBC-1、MA细胞凋亡.结论 ZGDHu-1能抑制A549、EBC-1、MA细胞增殖,并可诱导其细胞凋亡.
Objective To study the effect of N, N-di-( m-methylphenyi) -3,6-dimethyl-l ,4-di-hydro-1,2,4,5-tetrazine-1,4-dicarboamide (ZGDHu-1) on proliferation and apoptosis of A549, EBC-1 and MA cells in vitro. Methods Different concentrations of ZGDHu-1 were incubated with A549, EBC-1, or MA cells for different durations. The proliferation inhibition was analyzed by alive cell count, Methyl thi-azolyl tetrazolium (MTT) assay and BrdU-ELISA. Cell apoptosis was measured by cell morphology, DNA content, Annexin-V/PI and Hoechst33258 labeling method. Results ZGDHu-1 could inhibit proliferation of A549, EBC-1 and MA cells within a certain range of treating duration and does, with a 48 h IC50 of 165, 106, 210μLg/L, 72 h of 42, 18, 90μg/L, and a majority of A549, EBC-1 and MA cells were arrested in G2/M phase. The apoptosis of A549, EBC-1 and MA cells was confirmed by type cell morphology, DNA fragment, sub-G1 phase and Hoechst33258 and Annexin/PI labeling method. Conclusion ZGDHu-1 can inhibit proliferation and induce apoptosis of A549, EBC-1, and MA cells.
