采用反相高效液相色谱法分别对鲜人参、生晒参、红参中6种低分子量有机酸(乙酸、丙二酸、乳酸、苹果酸、柠檬酸、琥珀酸)进行分析。色谱柱为Kromasil C18(250 mm×4.6 mm,5μm),柱温30℃,5%甲醇-0.01 mol/L KH2PO4(p H 2.5)为流动相,流速1 m L/min,进样量20μL,检测波长210 nm。各有机酸在0.001 6~1.000 0 mg/m L内线性关系良好,6种有机酸的平均加样回收率为97.54%~102.53%。该方法准确、高效地测定了人参中的低分子量有机酸含量。
Six low molecular weight organic acids( acetic acid,malonic acid,lactic acid,malic acid,citric acid and succinic acidin) in fresh ginseng,white ginseng and red ginseng were determined by reversed phase high performance liquid chromatography( RP-HPLC). Kromasil C18column( 250 mm×4. 6 mm,5 μm) was used at 30 ℃ and mobile phase was 5% CH3OH-0. 01 mol / L K2HPO4( p H 2. 5) with flow rate of 1 m L/min and injection volume was 20 μL. Detection wavelength was 210 nm. Six organic acids had a good linearity in range of 0. 001 6-1. 000 0 mg / m L,and average recoveries of the six organic acids were in the range of 97. 54% to 102. 53%. This method has accurately and effectively determined low molecular weight organic acids in Panax ginseng.
