[目的]从基因组水平上阐明水稻在与白叶枯病菌(Xanthomonas oryzae pv. oryzae,Xoo)感病互作条件下基因表达反应的特征.[方法]用cDNA-AFLP方法对Xoo接种处理不同时间的水稻细胞进行基因表达谱分析.[结果]在测定的大约4 000个水稻cDNA片段中,363个(9.1%)是差异性表达的,其中295个受Xoo诱导上调表达,68个被抑制下调表达.根据在不同互作时间点的表达特征,它们可分为7种表达类型.对31个差异片段的序列同源性进行分析,发现它们在己公布的水稻全基因组中完全相同的序列,但只有10个基因具有已知或推断的生物学功能,其余21个基因功能未知.用荧光实时定量PCR分析法证实了cDNA-AFLP基因表达结果.[结论]构建了水稻-Xoo感病组合在细胞水平上互作的基因表达谱,从基因组水平上识别了一批受Xoo诱导或抑制表达、与水稻感病反应相关的基因,这些新基因的发现为开展水稻感病功能基因组学的研究提供了材料.
[ Objective ] The study was conducted to elucidate the gene expression response of rice to bacterial inoculation with Xanthomonas oryzae pv. oryzae (Xoo) at the genomic levels. [Method ] cDNA-amplified fragment length polymorphism (cDNA-AFLP) was employed to analyze the gene expression patterns of mock- and Xoo-inoculated rice suspension cultured cells. [ Result ] Three hundred and sixty-three (9.1%) from approximately 4 000 cDNA fragments analyzed were differentially regulated (295 up-regulated and 68 down-regulated) 0.5, 3, 24 h after co-cultivation with Xoo. Seven gene expression patterns of the differential fragments were identified. Ten of 31 sequenced fragments showed homology to genes with known or putative functions involved in metabolism, pathogen response and signaling. 21 others did not show any homology to sequences with known functions. cDNA-AFLP differential patterns for 5 selected genes were confirmed via real-time reverse transcription PCR analysis. [ Conclusion ] Gene expression profiling in response to Xoo and differentially expressed genes of rice were revealed and identified via cDNA-AFLP analysis. This work begins to reveal potential disease susceptibility-related genes of this staple crop with global importance.
