本研究在基于前期对拟南芥VDAC(电压依赖性阴离子通道蛋白质)与AtGluRS(谷氨酰tRNA合成酶)两种基因的过量表达和抑制表达转基因突变体株系的初步生理性状分析基础上,对筛选获得的转基因T1代植株以及拟南芥abil、abi2突变株,进行转录水平两种蛋白特异性基因片段的地高辛标记Northern杂交以及半定量RT—PCR分析.结果显示,在拟南芥abil、abi2突变株中,VDAC和AtGluRS两种基因的转录表达均收到不同程度的抑制;而VDAC与AtGluRS两种基因之间存在间接或直接的正调控关系,进一步证明了两种基因在转录水平上存在着密切联系.
Based on primitive physiological traits study lines of VDAC and AtGIuRS in Arabidopsis thaliana, of the over expression and suppression transgenic analyzed the mRNA transcription level of the selected T1 generation of the two transgenic lines and abil and abi2 mutants through Northern blot and semiquantitative RT-PCR of specific genes. It is revealed that the gene transcription of VDAC and AtGluRS had been inhibited inordinately in abil and abi2 mutants. And positive regulation was existed directly or indirectly between VDAC and AtGluRS as well, which had been proved the close relationship at the transcription level between VDAC and AtGluRS in Arabidopsis thaliana.
