蛋白酶抑制剂(Protein inhibitors,PIs)是一类小分子多肽或蛋白质,在植物抵抗生物和非生物逆境中起中起重要作用。利用PCR方法从强抗逆植物蒙古沙冬青(Ammopiptanthus mongolicus)中克隆到一个胰蛋白酶抑制剂(Trypsin inhibitors,TIs)基因AmTI,分析了其在非生物胁迫下的表达变化,构建了其植物表达载体。结果表明,AmTI的编码蛋白含197个氨基酸残基,分子质量为21.5kDa,在N端有信号肽序列,属于Kunitz型;在正常生长条件下,该基因只有微量的基础表达,但在脱水、低温和盐胁迫处理下,其表达量显著上调。本研究为进一步揭示AmTI的功能奠定了基础。
Protein inhibitors are a group of polypeptides or proteins and play important roles in the resistance of plants to biotic and abiotic stresses.Ammopiptanthus mongolicus is a plant with strong resistance to abiotic stresses.In this paper,a trypsin inhibitor(TI) gene,designated AmTI,was cloned from the plant by PCR method.Then expression profiles of the gene under some abiotic stresses was detected,and its plant expression vector was constructed.The results showed that the predicted protein encoded by AmTI consists of 197 amino acids and has a molecular mass of 21.5kDa and a N-terminal signal peptide.The protein was categorized into Kunit-type TI.In normal condition,AmTI expressed at a very low level,under dehydration,cold and salt treatments,however,its expression increased significantly.The study paved way for elucidating the gene function subsequently.
