鞘内注射Akt抑制剂Ⅳ对注射式根性神经痛大鼠痛行为学的影响

目的 探讨鞘内给予Akt抑制剂Ⅳ对注射式根性神经痛大鼠痛行为学的影响.方法 选择成功鞘内置管后无运动障碍的雄性SD大鼠18只,用分段随机分组法分为假手术组(S组)、溶剂对照组(C组)和Akt抑制剂组(A组),每组6只.C组和A组均采用注射surgiflo制备注射式根性神经痛模型,A组于造模前30 min给予Akt抑制剂Ⅳ0.6μg/10 μl.S组和C组则给予等量的10%酒精.各组均于造模前、造模后第1、4、7、10、17、28天测量双侧足底机械缩足阈值(paw witdrawal mechanical threshold,PWMT)和热辐射刺激潜伏期(paw withdrawal thermal latency,PWTL).结果 与S组相比,造模后第1天C组同侧肢体PWMT(9.56±1.49vs 14.03±0.94)和PWTL(9.91±1.80 vs 19.96±1.69)明显降低(P<0.05),持续至术后28 d;而对侧PWMT和PWTL与S组则无明显变化(P>0.05).与C组相比,A组痛行为学明显增加(PWMT:4.88±1.36 vs 7.51±0.88;PWTL:5.83±1.19 vs 8.19±1.66),并持续至术后7 d.结论 鞘内注射Akt抑制剂Ⅳ可以加重慢性背根神经节压迫引起的痛行为学,Akt在注射式根性神经痛大鼠痛行为学的形成中起重要作用.

Objective To investigate the effects of intrathecal injection of Akt inhibitor Ⅳ on pain behavior in rats model of injected radicular pain. Methods Eighteen male Sprague Dawley (SD) rats with successful intrathecal intubation were randomly divided into three groups (n=6): Sham group (S), injected radicular pain group (C), and Akt inhibitor group (A). In C group and A group, the model of injected radicular pain were established by surgiflo injection. Akt inhibitorⅣ was injected 30 min before the model preparation in group A. Vehicle (10% ethanol) was intrathecally injected at the same time in group C and group S. Pain ethology indexes such as paw withdrawal mechanical threshold (PWMT) and paw withdrawal thermal latency (PWTL) were assessed on 1 d before and 1, 4, 7, 10, 17 d and 28 d after operation. Results Compared to group S and preoperative level, PWMT and PWTL were decreased on the first postoperative day in group C (P〈0.05). Intrathecal Akt inhibitorⅣ injection significantly decreased PWMT and PWTL (P〈0.05) in groups A. Conclusion Intrathecal Akt inhibitorⅣ injection would intensify pain behavior in rat model of injected radicular pain. Akt may play an important role in the development of pain behavior in rat model of injected radicular pain.
