以分子量100D和500D规格的透析袋做工具,对吴茱萸和吴茱萸汤水提醇沉部分进行透析纯化,分别得于〈100D,100~500D以7L〉500D共3部分。利用高效液相色谱对不同时间透析处理过的部分化学成分进行分析,确定透析效率最佳时间点,并以此对血管活性选行体外实验。通过比较吴茱萸碱、异鼠李素和柠檬苦素3种成分在不同透析时间处理下100~500D区间内的含量,初步确定48h透析处理为最佳透析效率时间点。通过检测该时间吴茱萸汤处理组中6-姜酚、人参皂苷Re、人参皂苷Rg1和人参皂苷Rb1的含量,进一步验证48h为最佳处理时间。吴茱萸汤和吴茱萸〉100D透析部分具有明显的缩血管活性,〈100D透析部分对血管收缩无明显影响。表明利用不同孔径和透析袋对提取物进行样品前处理,并用于体外实验具有一定的可行性。
Dialysis tubes (Mw 100 D and Mw 500 D) were used to dialyze and purify the water extraction of Wuzhuyu (Fructus Evodiae) and Wuzhuyu decoction, and three portions of 〈100 D, 100-500 D and 〉500 D were obtained. To ascertain the peak efficiency timepoint, several chemicals treated for different time periods by HPLC were analyzed, and the activity of blood vessels was studied in vitro. By comparing different dialyzing time for three chemicals including evodiamine, isorhamnetin and limonin in 100-500D,the best treatment time was found to be 48 h. The portion of greater than 100 D significantly contracted blood vessels, while the portion of less than 100 D did not. In conclusion, it may be feasible to dispose samples using dialysis tubes with different apertures and apply the extract to pharmacological evaluation in vitro.
