目的探讨应激所致肠功能紊乱大鼠模型建立和评价的方法。方法 SD大鼠随机分为肠功能紊乱模型组和正常组,模型组采用慢性束缚+过度疲劳+饮食失调法造模,3周后取血,用酶联免疫法(ELISA)试剂盒在血清测GAS、IL-2,在血浆中测ACTH、MTL、SS。结果模型组大鼠3周后与正常组相比,外观行为发生明显变化,体重和肝脾重量显著降低,生物标志物水平降低。结论通过慢性束缚+过度疲劳+饮食失调法可以建立稳定的应激肠功能紊乱大鼠模型,GAS、IL-2、ACTH、MTL、SS等内源性物质可作为模型的评价指标。
Objective To explore the method of establishment of a rat model of stress-induced intestinal dysfunction and the method of its evaluation. Methods Sprague-Dawley rats were randomly assigned to two groups: the normal group and the model group. The model group was developed by combination of chronic constraint,excess fatigue and eating disorders for three weeks. At end of the third week,the serum GAS,IL-2,and plasma MTL,SS and ACTH were measured using an ELISA kit. Results Compared with the normal group,the model group had many distinct changes of appearance and behavior, weight loss, decrease of liver and spleen weight, and apparent changes of many biomarkers.Conclusions Stable rat model of stress-induced intestinal dysfunction can be successfully established by combination of chronic constraint,excess fatigue and eating disorders in SD rats,and the biomarkers such as GAS,IL-2,ACTH,MTL,SS can be used to evaluate this model.
