以巨大口蘑为试材,采用均匀设计法优化原种培养基配方,用数理统计软件DPS对试验结果进行二次多项式逐步回归分析,系统研究了蔗渣、棉籽壳、椰子壳、麸皮、石灰、轻质碳酸钙、硫酸镁、磷酸二氢钾8种成分对巨大口蘑菌丝生长速度和高度的影响。结果表明:原种培养基最优配方为蔗渣14.80%、棉籽壳42.44%、椰子壳35.80%、麸皮3.60%、石灰2.86%、轻质碳酸钙0.20%、硫酸镁0.20%、磷酸二氢钾0.10%,在此条件下巨大口蘑生长速度为1.32cm/d,显著高于筛选试验中最优配方3的生长速度0.36cm/d。
Taking Tricholoma giganteumas cultivation material,uniform design method was used to optimize the formula of culture medium for second-class spawn,quadratic polynomial step regression analysis method was employed for analyzing the experiment results.Eight components of bagasse,cotton seed hull,coconut shell,bran,lime,light Calcium carbonate,MgSO4,KH2PO4 were analyzed for the major characters including rate of mycelial growth and length of mycelial.The results showed that the optimum formula was bagasse 14.80%,cotton seed hull 42.44%,coconut shell35.80%,bran 3.60%,lime 2.86%,light CaSO40.20%,MgSO40.20%,KH2PO40.10%.Under this condition,the growth speed of Tricholoma giganteum was 1.32cm/d and it was significantly faster than the growth speed of formula3(0.36cm/d)in the screening experiments.
