从猴头菇(Hericium erinaceus)子实体中,经过除杂、粉碎、热水浸提、醇沉处理后,得到了猴头菇杂聚糖粗提物,并运用 DEAE-cellulose column 进行柱层析分离纯化,得到猴头菇寡聚糖(HEO-A).通过水解,单糖分析,扫描电镜技术(SEM ),高效凝胶渗透色谱(HPGPC),红外光谱技术(IR )和核磁共振技术(NMR)对猴头菇寡聚糖进行了初步的结构分析.结果显示,猴头菇寡聚糖由木糖和葡萄糖两种单糖构成,均重分子量为1877 D .在体外化学模拟条件下,研究了不同浓度的猴头菇寡聚糖对羟基自由基(·OH)、1,1-二苯基-2-苦苯肼自由基(DP-PH ·)及ABTS 自由基的清除作用.对DPPH ·自由基的清除能力,其IC50值为12.5 mg/mL ;对·OH自由基的清除能力,当 HEO-A 浓度达到5 mg/mL 时,其清除率可达到最大(37.51%);对ABTS 自由基的能力,其IC50值为0.93 mg/mL ,且当 HEO-A 的浓度为1.5 mg/mL时,清除率即可达95.44%.这些结果说明猴头菇寡聚糖(HEO-A)具有显著的抗氧化功能,可以作为一种理想的天然抗氧化剂资源.
A oligosaccharide was extracted from the fruiting bodies of Hericium erinaceus with water and then successively purified by DEAE-cellulose 52 column chromatography ,yielding one major oligosac-charide fractions :HEO-A .Structural features of HEO-A were investigated by a combination of total hydrolysis ,high-performance gel permeation chromatography analysis (HPGPC) ,scanning electron mi-croscope (SEM) ,infrared (IR) spectra and nuclear magnetic resonance (NMR) spectroscopy studies . The results showed that the average molecular sizes of HEO-A were approximately 1877 D and was mainly composed of xylose and glucose .The antioxidant activity analyses revealed that HEO-A pos-sessed considerable antioxidant activity .The IC50 value of the DPPH · radical scavenging activity is 12 .5 mg/mL .The · OH radical scavenging ability reaches the maximum (37 .51% ) when the concentration of HEO-A reached 5mg/mL .The IC50 value of the ABTS radical scavenging activity is 0 .93 mg/mL , and w hen the concentration of HEO-A is only 1 .5 mg/mL ,removal rate is up to 95 .44% .Accordingly , Hericium erinaceus might serve as an effective antioxidative healthcare food and source of natural an-tioxidants .
