目的:用CD40高表达细胞膜色谱模型筛选抗动脉粥样硬化中药丹参的有效成分。方法:构建CD40高表达的内皮细胞(ECV-304),制备细胞膜固定相,应用CD40高表达的内皮细胞膜色谱模型筛选丹参中抗动脉粥样硬化的有效成分。结果:丹参酯溶性和水提取部位均存在对CD40高表达细胞膜的亲和作用,而由丹参酯溶性和水提取部位中分离得到的成分丹参酮ⅡA、丹参酮ⅠA和丹参素则是其有效成分。结论:CD40高表达细胞膜色谱模型可用于抗动脉粥样硬化药物的筛选。
Objective: To screen the effective components of Salvia miltiorrhiza for atherosclerosis with chromatography of CD40 highly expressed cell membrane. Methods: The stable high expression of CD40 in human endothelial ceils ECV-304 was constructed, and the cell membrane stationary phase (CMSP) was prepared to screen the effective anti-atherosclerostic components of Salvia miltior- rhiza with it. Results: It was found that liposoluble and water soluble extracting site from Salvia miltiorrhiza had affinity for the cell membrane With highly expressed CD40, in which Tanshinone ⅡA, Tanshinone ⅠA and Danshensu were effective components. Conclusion: The model of the newly constructed cell membrane chromatograhy may be used to screen druas for atherosclerosis.
