目的:探讨内皮细胞(endothelial cells,ECS)对血管平滑肌细胞(vascular smooth muscle cells,VSMCs)整合素β1(β1-integrin)表达的影响,为研究血管壁细胞功能提供理论依据.方法:应用血管ECs VSMCs联合培养系统,以Western Blot和细胞免疫荧光技术,检测与ECs联合培养12h后VSMCs的β1-integrin表达和分布变化。结果:与ECs的联合培养,能明显增强VSMCs的β1-integrin蛋白表达,同时诱导VSMCs表面β1-integrin的激活。结论:ECs可能通过渊节VSMCs的β1-integrin表达,从而影响VSMCs的功能。
Objective: To study the effects of coculture with endothelial cells (ECs) on the expression of β1-integrin in vascular smooth muscle cells (VSMCs). Methods: Human umbilical veins ECs and VSMCs were isolated. For coculture studies, ECs were grown to confluent on one side of a porous polyethylene terephthalate (PET) membrane. VSMCs were then plated on the opposite side of the membrane and cocultured for 12 hours. Next, the expression of β1-integrin of VSMCs was measured by Western blot and immunofluorescencc microscope. Results: VSMCs cocultured with ECs exhibited a significant increase in the expression of β1-integrin comparing to that of the control, i.e., VSMCs cultured alone. EC-coculture resulted in increased β1-integrin localization to the attached surface of VSMCs. Conclusions: The results suggest that induction of the expression ofVSMC β1-integrin by ECs, may partially influence VSMC function.
