目的:建立中医阴黄证动物模型,系统评价茵陈四逆汤对大黄、乙醇、α-异硫氰酸萘酯(ANIT)三因素复合诱导的大鼠阴黄证的治疗作用。方法:70只Wistar大鼠随机分为7组,分别为空白组、模型组、模型恢复组、茵陈四逆汤低剂量组、茵陈四逆汤中剂量组、茵陈四逆汤高剂量组、阳性药对照组。观察大鼠一般生物学状态,检测谷丙转氨酶(ALT)、谷草转氨酶(AST)、碱性磷酸酶(ALP)、1-L-谷氨酰转肽酶(γ-GT)、总胆红素(TB)、直接胆红素(DB)、总胆汁酸(TAB)、血清白介素2(IL-2)的活性和效价以及环磷酸腺苷与环磷酸鸟苷比值(cAMP/cGMP),同时观察大鼠肝脏组织病理学情况。结果:阴黄证大鼠出现明显阳虚表现。模型组、模型恢复组ALT、AST、ALP、^y—GT、TB、DB、TAB、IL-2活性显著高于正常对照组(P〈0.01),治疗组上述指标,与模型组及模型恢复组比下降显著(P〈0.01);模型组、模型恢复组cAMP/cGMP比值显著低于正常对照组(P〈0.01),治疗组cAMP/cGMP比值与模型组及模型恢复组比升高显著(P〈0.01)。同时,组织病理学也得出上述相同的结论。结论:茵陈四逆汤对阴黄证大鼠具有良好的治疗作用。
This work aimed to investigate the effect of Yinchensini-Tang on Yin jaundice rats. 70 rats were randomly divided into 7 groups: the control group, the model group, the model recovery group, the Yinchensini-Tang low-dose group, the Yinchensini-Tang middle-dose group, the Yinchensini-Tang high-dose group, and the UDCA group. The changes of liver function and hepatic pathology and behavior were observed. The ALT, AST, ALP, γ- GT, TB, DB, TAB and IL-2 in the model group and model recovery group were significantly higher than in the control group (P〈0.01). The ALT, AST, ALP, γ-GT, TB, DB, TAB and IL-2 in the Yinchensini-Tang high-dose group were significantly lower than in the model group and model recovery group (P〈0.01), The cAMP/cGMP in the model group and model recovery group was significantly lower than in the control group (P〈0.01). The cAMP/cGMP in the Yinchensini-Tang high-dose group was significantly higher than in the model group and model recovery group (P〈0.01). The results showed that high-dosed Yinchensini-Tang could amend the changes of hepatic pathology in Yin jaundice rats. Thus Yinchensini-Tang has better effect on Yin jaundice rats.
