目的探讨不同浓度氟在不同时间段对三维培养体系中成纤维细胞增殖活性的影响。方法应用鼠尾胶原制备三维培养支架,种植小鼠皮肤成纤维细胞株(L929)。将细胞分为对照组和6个染氟组(0.001、0.002、0.1、1、10和20mg/LF^-),采用四唑盐(MTT)比色法,检测氟化物作用1、2、4、24、48、72和96h对成纤维细胞细胞增殖特性的作用。结果成纤维细胞在加氟初期仅个别组(1h和2h的0.002mg/L组和2h的0.1mg/L)细胞增殖活性有增加,其他组下降或明显下降;加氟24、48和72h时间段各加氟组成纤维细胞细胞增殖活力多高于未加氟组,未达统计学差异显著程度。结论氟对三维培养中的成纤维细胞的增殖活力有刺激作用,但作用发生较慢,程度相对较弱。
Objective To study the effects of proliferated activity of three - dimensional cultured fibroblasts in different dose and at different time. Methods The rat tail collagen were prepared the three - dimensional scaffold, fibroblasts ( L929 cell line) were cultured and planted in the three - dimensional scaffold, fibroblasts was divided into control group and 6 group of fluoride(0. 001,0. 002, 0.1, 1, 10and 20 mg/L F^-), MTT colorimetry was used to detect effects of fluoride on fibroblast proliferated activity after existence of fluoride 1,2, 4, 24, 48, 72 and 96 h. Results Fluoride increased the proliferated activity of fibroblasts in certain group( 1 h and 2 h 0. 002 mg/L, 2 h 0.1 mg/L). The proliferation of fibroblasts was increased in the initial stage as the existence of fluoride , the others was decreased or obviously decreased ; Fluoride increased the proliferated activity of fibroblasts than the control group at certain time (24, 48, 72 h) . but the results didn't reach the significant level of statistics. Conclusions Fluoride increased proliferated activity of the three - dimensional cultured fibroblasts , nevertheless, the effect was fairly slow, the degree was relatively feeble.
