Objective To establish a method for preparing the Dulbecco＇s Modified Eagle Medium：Nutrient Mixture F12（DMEM/F12）.Methods The related experimental materials must be sterilized.Add one bag of powdered medium DMEM/F12 and 1.2 g of NaHCO3 into 1 000 mL distilled water,after being stirred with magnetic stirring apparatus,DMEM/F12 solution were filtrated immediately with defecator connected with vacuum pump and preserved in 4℃ refrigerator about 6 months.DMEM/F12 supplemented with serum and something else will be maintained about 4 weeks.Results Prepared DMEM/F12 were not been polluted and good of cells culturing.Conclusion Sterilize the related materials strictly and prepare the culture medium DMEM/F12 aseptically is effective for culturing cells.
