目的:建立同时测定朝鲜淫羊藿中新绿原酸、绿原酸、隐绿原酸和金丝桃苷含量的超高效液相色谱(UPLC)方法。方法:采用Acquity BEH C18色谱柱(2.1mm×50mm,1.7μm);以0.1%乙酸-甲醇为流动相,梯度洗脱;流速:0.4mL/min;柱温:45℃;检测波长为325nm和353nm。结果:新绿原酸、绿原酸、隐绿原酸和金丝桃苷分别在0.576-57.600、1.806-180.600、1.056-105.600和1.530-153.000μg/mL内具有良好的线性关系(R2〉0.9996);平均回收率分别为96.54%,103.60%,98.61%和102.10%。结论:该方法测定朝鲜淫羊藿中新绿原酸、绿原酸、隐绿原酸和金丝桃苷的含量,具有简便快速、可靠的优点。
Objective: To develop an UPLC method for the simultaneous determination of neochlorogenic acid,chlorogenic acid,cryptochlorogenic acid and hyperin in Epimedium koreanum Nakai.Methods: Chromatography was performed on a Acquity BEH C18 column(2.1mm×50mm,1.7μm) with the mobile phase of methanol and water containing 0.1% acetic acid;Flow rate was 0.4 mL/min and column temperature was 45℃;The detection wavelength was set at 260nm and 325nm.Results: The correlation coefficient between concentration and chromatographic peak area of neochlorogenic acid,chlorogenic acid,cryptochlorogenic acid and hyperinwas over 0.9996 in the range of 0.576-57.600,1.806-180.600,1.056-105.600 and 1.530-153.000μg/mL,respectively.The average recoveries of the four compounds were 96.54%,103.60%,98.61% and 102.10%,respectively.Conclusion: The method is convenient and feasible for determination of the content of neochlorogenic acid,chlorogenic acid,cryptochlorogenic acid and hyperin in Epimedium koreanum Nakai.
