目的研究2型糖尿病鼠肝组织骨形成蛋白-9(bone morphogenetic proteins-9,BMP-9)基因转录及蛋白表达的变化。方法将大鼠随机分为2型糖尿病模型组与正常对照组,分别于造模后5、10、20 d处死,以RT-PCR及Western-blot对肝组织BMP-9基因转录及蛋白表达水平进行检测与分析。结果2型糖尿病模型组BMP-9基因转录及蛋白表达水平与正常组相比均显著性降低(P〈0.05);并且2型糖尿病大鼠5、10、20 d处死组BMP-9基因转录及蛋白表达水平呈逐步下降趋势。结论2型糖尿病大鼠肝组织BMP-9基因转录及蛋白表达水平较正常显著降低,这一变化可能与2型糖尿病的发生发展相关。
Objective To study changes of bone morphogenetic proteins (BMP) -9 gene transcription and protein expression in liver of type 2 diabetic rats. Methods Totally 20 Wistar rats were selected and randomly devided into type 2 diabetic model group and normal control group. Rats were killed at corresponding time point, and then BMP-9 gene transcription and protein expression in liver were detected by RT-PCR and Western-blot analysis. Results BMP-9 gene transcription and protein expression in type 2 diabetic model group were significantly lower than that in the normal control group (P 〈 0.05). Conclusion The BMP-9 gene transcription and protein expression in liver of type 2 diabetic rats are significantly lower. These changes may be associated with the production and progression of type 2 diabetes mellitus.
