目的观察土曲霉酮对人舌鳞状细胞癌细胞系SCC9的影响。方法不同浓度的土曲霉酮处理SCC9细胞后,用四甲基偶氮唑盐法检测细胞活力,用Annexin V/PI试剂盒及碘化丙啶分别检测细胞凋亡及细胞周期情况。结果土曲霉酮对SCC9细胞有抑制增殖的作用,抑制作用随着药物浓度的增大(P〈0.05)及给药时间的延长而增强(P〈0.05)。此外,与不给药组相比,5μmol/L和10μmol/L土曲霉酮处理,可引起处于G2/M期的SCC9细胞比例由(24.8±0.6)%分别增加到(26.2±0.4)%和(34.2±1.2)%(F=132.13,P〈0.01),但对SCC9细胞凋亡没有明显的影响。结论土曲霉酮通过引起G2/M期阻滞来抑制人舌鳞状细胞癌细胞系SCC9细胞的增殖,提示土曲霉酮有望在舌鳞状细胞癌的治疗中发挥作用。
Objective To study the effects of terrein on human tongue squamous cell carcinoma cell line SCC9. Methods SCC9 cells were treated with different concentrations of terrein for different time, then cell viability was meas- ured by MTF assay and tile cell cycle profile and apoptosis rate were analyzed by flow eytometry. Results Terrein inhib- ited the proliferatinn of SCC9 cells in a dose-depemlent and time-dependent manner. Compared to control group, 5 μmol/ L and 10 μmol/L terrein increased the cell proportion of G2/M phase from (24.8 ±0.61 )% to (26.2 ±0.40)% anti (34.2±1.16)% ( F = 132. 13, P 〈 0. 01 ), respectively. Conclusion Te,'rein inhibited the proliferation of SCC9 cells by inducing G2/M phase arrest.
