为探讨火炬松树干形成层内源激素分离提取的优化流程,以火炬松为材料,针对火炬松树干形成层富含松节油和色素等杂质的特点,通过使用不同的提取剂,如乙酸乙酯、石油醚、正己烷和浓度分别为10%、20%、30%、40%、50%、80%的甲醇提取,经高效毛细管电泳检测,确立了以30%的甲醇为提取液的火炬松树干形成层内源激素的提取优化流程,并利用该流程对不同倾斜角度的火炬松内源激素进行提取测定。结果表明,该方法可以同时分离提取测定5种内源激素,如脱落酸(ABA)、吲哚-3-丁酸(IBA)、玉米素(ZT)、吲哚-3-乙酸(IAA)和赤霉素(GA),平均回收率分别为100.02%、95.23%、96.56%、101.13%和103.12%
To optimize the extraction process of endogenous hormones from stem cambium of Pinus taeda by eliminating the turpentine and pigment etc.,effects of different extracting solvents like ethyl acetate,petroleum ether,normal hexane and methanol(concentrations of 10%,20%,30%,40%,50%,80%) on yield of endogenous hormones in stem cambium of Pinus taeda were investigated based on previous studies.Determined by high-performance capillary electrophoresis(HPCE),the optimum extraction process with extraction solvent of 30% methanol was obtained.In addition,the content of each endogenous hormone in the cambium of Pinus taeda was determined under the optimal extraction process.The method established in this study is favorable for the simultaneous extraction of ABA,IBA,ZT,IAA and GA in the stem cambium of Pinus taeda with mean recovery of 100.02%,95.23%,96.56%,101.13% and 103.12% respectively
