应用毛细管电泳/前沿分析法测定对硝基苯胺与牛血清白蛋白的相互作用结合常数。以对硝基苯胺与BSA的平衡混合溶液直接进样,根据对硝基苯胺的峰高求出游离对硝基苯胺的浓度,分别利用Scatchard方程和Klotz方程求解两者的相互作用结合常数和结合位点数。两种方法求得的结合常数分别是2 278.47 l/mol、2 413.95 l/mol,结合位点数分别是2.32、2.09,具有较好的一致性。讨论了毛细管电泳/前沿分析法在求结合常数方面的优越性。
Capillary electrophoresis/frontal analysis method was applied to study the interaction of the p-nitroaniline and bovine serum albumin to make a sample by combining with p-nitroaniline and BSA. The unbound concentration of p-nitroaniline can be determined from the plateau peak height of p-nitroaniline. The binding parameters were processed with both scatchard linear regrossion analysis and Klotz linear regrossion analysis. The binding constants are 2278.47 L/mol,2413.95 L/mol the binding sites are 2.32,2.09 respectively according to two linear regression analysis methods. The advantages of CE/FA in the study of protein-pollutant binding were discussed.
