目的:研究血府逐瘀汤含药血清对大鼠肾小球系膜细胞(RMC)经ox-LDL刺激后肿瘤坏死因子-α(TNF-α)和血凝素样氧化低密度脂蛋白受体-1(LOX-1)m RNA表达的影响,探讨血府逐瘀汤对RMC的保护性作用。方法:使用50μg/m L ox-LDL刺激RMC 24h,用包括不同浓度血府逐瘀汤、辛伐他汀含药血清进行干预24h,使用荧光定量PCR(RT-PCR)检测RMC中TNF-α和LOX-1的m RNA表达。结果:与模型组相比,不同剂量血府逐瘀汤含药血清和辛伐他汀含药血清均可不同程度地降低TNF-α及LOX-1的m RNA表达(P〈0.01),血府逐瘀汤含药血清的干预作用随浓度升高而升高,高浓度血府逐瘀汤含药血清的干预作用较辛伐他汀含药血清强(P〈0.01)。结论:血府逐瘀汤含药血清可减少RMC中TNF-α、LOX-1的表达,减轻ox-LDL对RMC的损伤作用。
Objective: To investigate the effects of serum containing Xuefu Zhuyu Decoction on expression of TNF- α and LOX-1 mRNA in rat mesangiat cells (RMC) intervened by ox-LDL, and to explore the protective effects of Xuefu Zhuyu Decoction on rat mesangial cells. Methods: RMC was intervened 24h by using 50μg/mL ox-LDL, and treated with serum containing Xuefu Zhuyu Decoction with different concentration and Simvastatin for 24h. RT-RCP was used to test the mRNA expression of TNF- α and LOX-1. Results: Compared with the model group, serum containing Xuefu Zhuyu Decoction with different concentration and S imvastatin could reduced the mRNA expression of TNF-α and LOX-1 at different degrees (P〈0.01), and the intervention effects of serum containing Xuefu Zhuyu Decoction increased with the increasing of concentration. The intervention effect of high concentration of serum containing Xuefu Zhuyu Decoction was stronger than serum containing Simvastatin (P〈0.01). Conclusion: Serum containing Xuefu Zhuyu Decoction could reduce the mRNA expression of TNF- α and LOX-1 and the damage in RMC caused by ox-LDL.
