目的主要观察新风胶囊(黄芪、薏苡仁、雷公藤、蜈蚣)对骨关节炎大鼠外周血免疫球蛋白IgG1、IgG2α及细胞因子IL-4、TNF-α,软骨、胸腺、脾脏自噬基因(Atg5、Atg7、Atg12)的表达的影响。方法大鼠随机设正常对照组和模型对照组(均予生理盐水)、氨基葡萄糖对照组,以及新风胶囊组,每组10只。除正常对照组外,采用关节腔注射木瓜蛋白酶和L-半胱氨酸方法制成骨关节炎大鼠模型,造模后第14天给药30 d。以光镜对大鼠软骨进行Mankin标准评分;采用ELISA法检测血清细胞因子IgG1、IgG2α、IL-4,和TNF-α;采用RT-PCR对骨关节炎大鼠软骨、胸腺、脾脏中的自噬相关基因Atg5、Atg7和Atg12进行检测。结果给药后,与正常组比较,模型组体质量明显下降,Mankin评分明显升高,血清IgG1、IgG2α、TNF-α升高,IL-4降低,在胸腺、脾脏、软骨等各组织中,Atg存在不同程度的降低(P〈0.05或P〈0.01);与氨基葡萄糖组比较,新风胶囊组体质量明显升高,Mankin评分明显降低,IL-4及Atg5、Atg7、Atg12升高(P〈0.01或P〈0.05)。结论新风胶囊可以抑制炎症反应,调节大鼠整体自噬水平,达到治疗骨关节炎。
AIM To observe the effects of Xinfeng Capsules( Astragali Radix,Coicis Semen,Tripterygium wilfordii Hook. f.,Scolopendrap) on the expressions of IgG1,IgG2α,cytokines IL-4,and TNF-α in peripheral blood and autophagy genes( Atg5,Atg7,Atg12) in cartilage,thymus and spleen in rats with osteoarthritis.METHODS Rats were randomly divided into the normal control group,the model group,glucosamine group,and Xinfeng Capsules group( XFC),10 rats in each group. Except the normal control group,rats with osteoarthritis were prepared by intraarticular injection of papain and L-cysteine; the administration lasted for thirty days from the fourteenth day of modelling; the normal control group and model group were given saline; the other two groups were given glucosamine and XFC suspension,respectively. Mankin standard score for rat cartilage was obtained by light microscopy; ELISA method was used to detect cytokines IgG1,IgG2α,IL-4,and TNF-α in serum. The gene analyses of Atg5,Atg7 and Atg12 in cartilage,thymus and spleen were detected by RT-PCR. RESULTS After the administration,compared with the normal control group,the body mass of the model group significantly decreased; Mankin score markedly increased; so did serum IgG1,IgG2α,and TNF-α; IL-4 decreased. The existance of Atg decreased to some extent in the tissues of thymus and spleen and cartilage( P〈0. 05 or P〈0. 01). As compared with glucosamine group,the body mass of XFC group increased; Mankin score significantly decreased; IL-4 and Atg increased( P〈0. 01 or P〈0. 05). CONCLUSION Xinfeng Capsules can treat osteoarthritis through both inhibiting inflammation and adjusting the overall autophagy levels in rats.
