以番茄品种辽园多丽离体花柄为试材,分别在25(CK),30,35℃下进行番茄花柄外植体培养,研究不同温度处理对番茄离体花柄脱落过程及其相关酶活性的影响。结果表明:在25~35℃培养条件下,番茄离体花柄的脱落率随温度升高而有不同程度降低,其中经32h培养,35℃处理未发生脱落,而25℃和30℃处理的脱落率分别为24-32%和12.39%。35℃处理花柄离区可溶性蛋白含量随处理时间延长迅速降低,而25℃和30℃处理变化不大。35℃处理明显降低了离区纤维素酶与多聚半乳糖醛酸酶的活性.30℃处理纤维素酶活性略有升高,而多聚半乳糖醛酸酶活性在处理32h后显著下降。35℃亚高温抑制番茄离体花柄脱落可能是与纤维素酶和多聚半乳糖醛酸酶活性,特别是多聚半乳糖醛酸酶活性受到抑制有关。
In this paper, effects of different temperatures on pedicel abscission in vitro and abscission-related enzymes in tomato were studied to make clear the effects of sub-high temperatures on abscission process. The pedicels of 'Liaoyuanduoli' tomatoes were cultured in vitro under 25, 30, 35℃, respectively. The abscission rate reduced with the rising of temperature. The abscission rate was zero under 35℃ for 32h, but it was 24.32% and 12.39% under 25℃ and 30℃. The content of soluble protein in abscission zone decreased under 35℃ treatment, but there was no significant difference between 25℃ and 30℃ treatments. In addition, activities of cellulase and polygalacturonase in abscission zone obviously decreased under 35℃ treatment, while cellulase activities slightly increased and polygalacturonase activities substantially decreased under 30℃ treatment after 32 h. The results indicated that the inhibition of abscission process under 35℃ treatment was due to the low level of cellulase and polygalactur- onase activities, especially the latter under sub-high temperature.
