目的研究肉桂对滋肾丸中盐酸小檗碱在正常大鼠前列腺分布动力学的影响。方法将sD大鼠分为滋肾丸组(31.5s/kg)与知柏组(30.0g/kg)2组,每组25只,灌胃水煎液一次,药后1、3、4、6、14h,腹主动脉取血,摘取前列腺。采用C-18固相萃取柱处理样品,用HPLC法测定血浆和前列腺组织中盐酸小檗碱的含量。色谱条件:流动相乙腈-0.01mol磷酸二氢钾(内含0.2%SDS)(43:57);流速0.8mL/min;检测波长345nna。结臬盐酸小檗碱在血浆中的线性范围为0.6—20ng/mL,最低检测限为0.3ng/ml,日内及日间RSD分别为5.77%与14.24%,平均回收率为98.87%;盐酸小檗碱在前列腺组织中的线性范围为1.25,100ng/mL,最低检测限为0.625n-ml,日内与日间RSD分别为8.7%与10.68%,平均回收率为97.21%。滋肾丸和知柏中盐酸小檗碱在前列腺中的分布系数分别为3,45与2.47;与知柏组比,盐酸小檗碱在前列腺中的相对摄取率为1.08,峰浓度比为2.10;滋肾丸组血浆盐酸小檗碱的c-明显降低(P(0.05),前列腺盐酸小檗碱的c。明显升高(P〈0.05)。结论盐酸小檗碱在前列腺的渗透性较强,肉桂增强盐酸小檗碱在前列腺的渗透性可能是其引经作用的机制之一。
bjective To explore effects of cassia on distribution pharmacokinetics of berberine hydrochloride of Zishen Pill in rats' prostate.Methods SD rats were divided into a ZishenPill group (31.5 g/kg) and a Zhibai group (30.0 g/kg), 25 cases in each one. Intragastric administration of water decoction was given once. lh, 3h, 4h, 6h and 14h after treatment, blood in abdominal aorta and prostate were collected. The C-18 solid phase column was applied to analyze samples, and HPLC method was to measure level of berherine hydrochloride in plasma and prostate tissue.Chromatographic condition included mobile phase of acetonitrile-0.01mol potassium dihydrogen phosphate (43:57), flow speedof 0.SmL/minand wavelength of 345nm. Results The linear range of berberine hydrochloride in plasma was 0.6-20 ng/ml; the lowest detectable limit was 0.3ng/mL; the RSD of within-day and between-day precision was 5.77 % and 14.24% respectively; the average recovery was 98.87 %. The linear range of berberine hydroehloride in prostate tissue was 1.25-100 ng/mL; the lowest detectable limit was 0.625 ng/mL; the RSD of within-day and between-day precision was 8.7 % and 10.68 % respectively; the average recovery was 97.21% .The distribution coefficient of berberine hydroehloride inZishen pill and Zhibaiwas 3.45 and 2.47 respectively.Compare with Zhibai group,the relative uptake rate and peak concentration ratio of berberine hydroehloride in prostate tissue was 1.08 and 2.10 respectively; the Cmax of berberine hydrochloride in plasma in ZishenPill group was decreased significantly (P〈0.05), while thatin theprostate tissue was increased significantly (P〈0.05).Conclusion The berberine hydrochloride has higherpenetrability in prostate. Cassia increasing penetrability of berberine hydroehloridein prostate may be one of meridian guiding effect mechanism.
