目的:研究骨硬化素单链抗体对去卵巢模型大鼠骨质疏松性骨折愈合的影响。方法:取24只大鼠,手术切除卵巢后l周,用开放手采建立左下肢股骨中段横行骨折模型,随机分为模型组和治疗组,每组12只,模型组犬鼠鞘内注射磷酸盐缓冲液O.2ml,治疗组大鼠鞘内注射骨硬化素单链抗体2.5mg/kg,每周2次。连续给药12周后,检测各组大鼠血清中钙、磷、碱性磷酸酶、骨钙素舍量,左侧股骨的骨密度(BMD)、骨小梁厚度(Tb.Th)、骨小梁间距(Tb.Sp)、骨体积分数(BVF)、结构模型指数(SMI)和形态学变化。结果:模型组大鼠血清中钙、磷、碱性磷酸酶、骨钙素含量分别为(1.92±0.54)mmolfL、(1.54±O.61)mmol/L、【64.43±13.92)U/L、(9.04±0.71)ng/ml,治疗组大鼠上述指标含量分别为(2.43±0.28)mmol/L、(1.58±0.26)mmol/L、(79.25±12.49)U/L、(9.57±O.83)ng/ml;模型组大鼠BMD、Tb.Th、Tb.Sp、BVF、SMl分别为(91.51±12.33)mg/cc、(O.09±O.02)mm(O.47±O.22)mm、0.18±0.03、1.42±O.31.治疗组大鼠上述指标分别为(135.15±11.12)mg/cc、(O.01±O.03)toni、(0.34±0.11)mm、0.29±0.01、O.65±O.34。与模型组比较,治疗组大鼠血清中钙、碱性磷酸酶含量和BMD、BVF均明显增加(P〈O.05),SMI明显减小(P〈0.05),其余指标差异无统计学意义(P〉0.05)。镜下观察治疗组大鼠成骨细胞数量比模型组多。结论:骨硬化素单链抗体对去卵巢模型大鼠骨质疏松性骨折愈合有促进作用。
OBJECTIVE: To study the impact of sclerostin single chain Fv on osteoporotic fracture healing in ovariectomized rats. METHODS: 24 SD rats were ovariectomized. One week later, the middle shaft transverse fractures model of the femur of left lower extremity was established. Model rats were randomly divided into model group and treatment group with 12 rats in each group. Model group was given intrathecal injection of phosphate buffer 0.2 ml, and treatment group was given intrathecal injection of sclerostin single'chain Fv 2.5 mg/kg, twice a week, for consecutive 12 weeks. The contents of calcium, phosphorus, alkaline phosphatise and osteocalcin in serum were determined. BMD, Tb.Th, Tb.Sp, BVF, SMI and morphologic change of left femur were also determined. RESULTS: The contents of calcium, phosphorus, alkaline phosphatise and osteocalcin in serum of model group were ( 1.92 ± 0.54) mmol/L, ( 1.54 ± 0.61) mmol/L, (64.43 ± 13.92) U/L and (9.04 ± 0.71 ) ng/ml; above index of treatment group were (2.43 ± 0.28) mmol/L, (1.58 ± 0.26)retool/L, (79.25 ± 12.49) U/L and(9.57 ± 0.83) ng/ml. BMD, Tb.Th, Tb.Sp, BVF and SMI of model group were (91.51 ± 12.33) mg/cc, (0.09 ± 0.02) mm, (0.47 ± 0.22) mm, 0.18 ± 0.03 and 1.42 ± 0.31 ; above in- dex of treatment group were ( 135.15 ± 11.12 ) mg/cc, (0.01 ± 0.03) mm, (0.34 ± 0.11) ram, 0.29 ± 0.01 and 0.65 ± 0.34, respective- ly. Compared with model group, the contents of calcium and alkaline phosphatise, BMD and BVF were increased significantly in treatment group (P〈0.05), and SMI was decreased significantly (P〈0.05) ; there was no statistical significance in other index (P〉0.05). The amount of osteoblast in treatment group was more than in model group under microscope. CONCLUSIONS: sclerostin single chain Fv promotes osteoporotic fracture healing in ovariectomized rats.
