以光色素为荧光探针,将其标记到萘抗体上,采用直接荧光免疫分析的方法,对土壤环境中的萘(NA)进行了检测。优化了试验条件,选择洗涤液PBST的pH值为7.40,温育时间为2h,抗原浓度为40.0μg/ml,标记抗体的稀释比为1∶20为最佳的实验条件。在此条件下,得到萘的标准工作曲线。线性范围是1.00×10-7~1.00×10-2 mg/ml,最低检出限为5.03×10-8mg/ml。并将这种方法检测了三种土样(即处理过生活污水、电镀废水以及印染废水的活性污泥)中的萘,回收率在97.53%~107.6%之间,结果令人满意。
We established a direct competitive immune-fluorescence method to determinate Naphthalene(NA) in the environment,using the Lumichrome as fluorescent probe.After optimizing the conditions,We chose pH 7.40,2 hours of reacting time,the concentration of antigen with 40.0μg /ml and the dilution rate of antibody with 1∶20 as the best conditions,then to change the concentration of NA for the experiment.It was found that a linear relationship between the logarithm of the concentration of NA and the restraint rate of reaction.The linear calibration range for the determination of NA was 1.00 × 10-7 ~ 1.00 × 10-2 mg /ml,and the linear equation was y = 7.505C + 61.54,R2 = 0.9917.The detection limit of this method was 5.03 × 10-8 mg /ml.By using this method,NA was determined in the soil from the living sludge,the plating sludge and the printing sludge.The results,with the recovery rate between 97.53% and 107.6%,were satisfying.
