目的从离体回肠运动的角度,探讨中药"十八反"中甘草与大戟、甘遂、芫花存在配伍禁忌的科学依据。方法制作大鼠离体回肠模型,以回肠收缩张力为指标,观察不同浓度的甘草与大戟、甘遂、芫花的单煎液、合并液、合煎液对大鼠离体回肠运动的影响。结果甘草单煎液呈现抑制回肠收缩作用,而大戟、甘遂、芫花各单煎液则表现为不同程度促进回肠收缩作用;甘草分别与大戟、甘遂、芫花合并给药后,可减弱后者促进回肠收缩的作用;各合煎液促进回肠收缩的作用均弱于其单煎液,其中甘草-甘遂合煎液的作用弱于其合并液,而甘草-大戟合煎液与甘草-芫花合煎液强于其合并液。结论甘草与大戟、甘遂、芫花合用后能显著抑制后者促进回肠收缩作用,从该角度探讨和揭示了甘草与具有峻下逐水功效的各药物存在配伍禁忌的作用特点与机制。
OBJECTIVE To find out the scientific basis of existing incompatibility of Glycyrrhizae Radix et Rhizoma with Euphorbiae Pekinensis Radix, Kansui Radix or Genkwa Flos among Eighteen Incompatible Medicament of TCM . METHODS The effects of single decoction, mixed decoction (mixed water extract of each individual herbs) and co-decoction (water extract of mixed two herbs) at different concentrations were observed with the index of rat ileal smooth muscle tension. RESULTS Single Glycyrrhizae Radix et Rhizorna decoction could inhibit the contraction of rat ileum, while each single decoction of Euphorbiae Pekinensis Radix, Kansui Radix or Genkwa Flos could promote the ileal contraction to different degrees. The effect of Euphorbiae Pekinensis Radix, Kansui Radix or Genkwa Flos would be weakened when it was mixed with Glycyrrhizae Radix et Rhizoma. The contraction-promoting effect of each co-decoction was weaker than the single decoction. And the effect of Glycyrrhizae Radix et Rhizoma-Kansui Radix co-decoction was weaker than the mixed decoction, whereas the effects of another two co-decoctions were stronger than the mixed decoction. CONCLUSION The contraction-promoting effect of Euphorbiae Pekinensis Radix, Kansui Radix or Genkwa Flos was markedly weakened when combined with Glycyrrhizae Radix et Rhizorna, And from this perspective, the characteristics and mechanism of imcompatiblity between Glycyrrhizae Radix et Rhizoma combined with drugs with purgative and eliminating fluid effects can be explored and indicated.
