目的研究云南省晋宁县彝族人群单核细胞趋化蛋白-1(MCP-1)G-2518A基因多态性分布的情况,为探讨MCP-1基因相关疾病的发病机制奠定基础.方法采用PCR-RFLP技术检测居住在云南省晋宁县310例彝族人群的MCP-1 G-2518A基因A/G多态性,计算AA、GA、GG基因型频率和A、G等位基因频率.结果云南省晋宁县彝族人群的MCP-1 G-2518A基因的AA、GA、GG基因型频率分别为20.0%、49.7%和30.3%,A、G等位基因频率分别为44.8%和55.2%.和不同种族人群相比较,云南晋宁县彝族人群的MCP-1 G-2518A基因A/G多态性分布与波兰人、印度人、韩国人、日本人、巴西人、欧洲人、美国人、土耳其人、德国人和希腊人等差异有统计学意义(P〈0.05),与中国香港人、中国汉人和台湾人等差异无统计学意义(P〉0.05).结论MCP-1 G-2518A基因型分布和基因频率存在种族和民族差异.
Objective To investigate the distribution of MCP- 1- 2518 gene A/G polymorphism in a Yi nationality population of Yunnan province, and to found the basis of pathogenesis about many diseases associated with it.Methods We recruited 310 healthy subjects of Yi nationality in rural area of Jinning, Yunnan province. The MCP- 1- 2518 gene A/G polymorphism was determined by PCR- RFLP technique. Frequencies for the AA, GA,GG genotypes and allele frequencies for the A, G alleles were calculated.Results In healthy people of Yunnan province, frequencies for the AA, GA and GG gene types were 20.0%, 49.7% and 30.3%.Derived allele frequencies for the A and G allele were 44.8% and 55.2%.Compared with the data from other countries, the distribution of MCP- 1- 2518 gene A/G genotypes and the allele frequencies in Yunnan Yi people differed significantly from that in Pole, Indian, Korean, Japanese, Brazilian, European, American, Turkish, German and Greek( P〉0.05), while did not differ significantly from that in Han Chinese, Taiwanese and people in Hong Kong( P〈0.05). Conclusion The frequency of genotype or the allele of MCP- 1- 2518 A/G polymorphism is different significantly in different ethnic populations and national populations.
