本文采用15%KOH的醇溶液除去小鼠单核巨噬细胞RAW264.7样品中的甘油三酯,用6%三氯乙酸除去其中蛋白质,然后以体积比为3:2的正己烷与异丙醇混合溶剂提取巨噬细胞内的胆固醇。采用硬脂酸键合硅胶色谱柱,以乙腈和异丙醇的混合溶剂为流动相,在检测波长212nm下测定胆固醇的含量。结果表明,胆固醇含量在0.005~0.3g/L之间时具有较好的线性关系,相对标准偏差为3.4%,检出限为1.5mg/L,回收率为96.04%。方法用于测定巨噬细胞中胆固醇的含量,结果满意。
An equal volume of freshly prepared KOH in ethanol (150 g/L) was added to monocyte-derived macrophages to hydrolyze the triglyceride, and 6% trichloroacetic acid was added to remove the protein. And then, the cholesterol of macrophages was extracted with hexane-isopropanol (3: 2, V/V). The cholesterol was analyzed using high performance liquid chromatography (HPLC). The C18 alkylamide silica column (150×4.6 mm, 5 μm) was selected and the eluent consisted of isopropanolacetonitrile 30:70(V/V) at a flow rate of 1. 0 mL/min. The detection wavelength was 212 nm. The calibration curve is linear in the range of 0.005-0.3 g/L for cholesterol (r=0. 9996). The detection limit is 1.5 mg/ L, relative standard deviation (RSD) is 3.4%, and the value of recovery is 96.04%. The method is rapid, accurate and reliable.
