【目的】建立龙血竭中新化合物一剑叶龙血素C的分离与测定含量方法。【方法】采用Phenomenex C18(250mm×4.6mm,5μm)色谱柱,甲醇-水(75:25,v/v)为流动相,流速为1m/min,检测波长为295nm,柱温50℃;用外标法定量。【结果】剑叶龙血素C在27.25—327ng范围有良好线性关系,r=0.9997,理论塔板数以剑叶龙血素C计为4500;方法回收率为96.90%,RSD为1.2%。【结论】本方法是测定龙血竭中剑叶龙血素含量的准确的定量方法。
[Objective] To establish a HPLC method for the separation and determination of Cochinchinenin C in Dragon' s Blood. [Methods] The chromatographic system consisted of Phenomenex C18 (250 mm×4.6mm,5μm) column and mobile phase of CH3OH-H2O (75 : 25) and detected at 295 nm, the temperature of column was 50℃ and quantitating with external standard method. [Results] The standard curves of Cochinchinenin C was linear in the concentration range of 27.25 - 327 ng ( r = 0.9997). The theoretical plate number is 4500. The average recovery rate was 96.90% (RSD =1.2%). [Conclusions] The established method is accurate, sensitive, simple and reliable for determination of Cochinchinenin C in Dragon's Blood.
