目的观察β-淀粉样蛋白1-40(Aβ1-40)大鼠海马双侧注射后大脑组织Caspase-3表达和海马神经细胞超微结构的变化及加减地黄饮子的干预作用。方法采用大鼠海马立体定向注射凝聚态Aβ1-40诱导阿尔茨海默病(AD)动物模型。采用免疫印迹方法检测大脑组织Caspase-3的表达,通过透射电子显微镜观察海马组织神经细胞超微结构的变化。结果模型组大鼠大脑组织Caspase-3蛋白表达相对值(2.647±0.6036)较假手术组(0.901±0.102)明显升高(P〈0.05),而加减地黄饮子(1.331±0.1053)可下调Caspase-3的表达(P〈0.05)。Aβ1-40可致模型组大鼠海马神经元的胞体质膜下出现大量的脂褐素,游离核蛋白体减少,加减地黄饮子对损伤有不同程度的恢复。结论加减地黄饮子通过上调Caspose-3蛋白的表达而发挥保护Aβ对大鼠脑神经细胞的损伤。
Objective To observe the effects of modified decoction of Rehmanniae on expression of caspase-3 protein and ultrastructural changes of pyramidal neurons in hippocampal in Alzheimer's disease (AD) rats induced by amyloid-β1-40 injection into brain. Methods AD model rats were induced by amyloid-β1-40 sterotaxis injectiorh Modified decoction of Rehmanniae was given by oral administration for 28 days. Expression of caspase-3 protein in rat brain was estimated by Western blotting method. Ultrastructure of pyramidal neurons in hippoeampal was observed by transmission electron microscope. Results The expression of caspase-3 in model group (2. 647 ± 0. 603 6) was significantly higher than that in sham group(0. 901 ± 0. 102 ) (P 〈 0.05 ), and that in the modified decoctioff Of Rehmanniae group (1. 331 ± 0. 105 3 ) was significantly lower than that in model group ( P 〈 0. 05 ). Lipofuscin deposit in hippocampal neurons in model group and the cell apparatus numbers decreased. Modified decoction of Rehmanniae treatment had protective effects on the neuronal cells damnification. Conclusions Modified decoction of Rehmanniae protect brain neurons from being injuried by Aβ1-40 by up-regulating the expression of caspase-3 protein.
