鞣酸对草乌中双酯型生物碱水解的影响

中文摘要:

目的:研究鞣酸对双酯型生物碱水解的影响,了解鞣酸与草乌模拟炮制情况。方法:采用HPLC法测定鞣酸模拟炮制品、水泡品及二者水煎液中苯甲酸、苯甲酰乌头原碱和乌头碱的含量,探讨不同模拟炮制品中双酯型生物碱的水解规律以及煎煮过程对双酯型生物碱水解的影响。结果:经模拟炮制后,鞣酸炮制品与水泡品中乌头碱的含量分别降低为原来的93%和60%;煎煮后,乌头碱继续水解,鞣酸炮制品和水泡品乌头碱分别水解了68%和98%;鞣酸炮制品水煎液中可检出苯甲酰乌头原碱,水泡品水煎液中可检出苯甲酸。结论:鞣酸可抑制双酯型生物碱在炮制及煎煮过程中的水解反应,且对一级水解和二级水解均有抑制作用。

英文摘要:

Objective: To explore the effect of tannin on hydrolysis of diester type alkaloid, and to learn the simulate processed of tannin and radix aconiti kusnezoffii. Methods: The HPLC method was used to determine the contents of benzoic acid, benzoyl aconine, and aconitine in samples of aconitum processed with tannin, aconitum processed with water, decoctions of aconitum processed with tannin and decoctions of aconitum processed with water, to discuss the hydrolysis of aconitum in different processed products, and the effect of their decoctions process on hydrolysis of diester type alkaloid. Results: The content of aconitum in samples of aconitum processed with tannin and processed with water were decreased with 93% and 60%; The content of aconitum in decoctions of aconitum processed with tannin and processed with water were continue to decrease with 68% and 98%; benzoyl aconine can be detected in decoction of aconitum processed with tannin, and benzoic acid can be detected in decoctions of aconitum processed with water. Conclusion: Tannin can inhibit the hydrolysis of aconitum during processing and decoction; The effect of inhibition were happened in both the first order hydrolysis reaction and the second order hydrolysis reaction.
