目的探讨骨髓间充质干细胞(MSCs)在皮肤创伤刺激下,能否从骨髓迁移到皮肤创面。方法抽取小型香猪的骨髓,分离纯化MSCs,培养扩增后,经DAPI标记回植到骨髓中,制作皮肤创面,7,14d,观察皮肤组织中是否有阳性细胞以及实验组和对照组的差别。结果将MSCs经DAPI标记回植后,实验组7dDAPI染色阳性的细胞较多,14d阳性细胞减少。而对照组阳性细胞稀少,在7,14d的皮肤组织切片中无差别。结论MSCs在皮肤创伤刺激下,可以从骨髓迁移到皮肤创面。
Objective To investigate the feasibility of bone marrow mesenchymal stem cells (MSCs) migrate from bone marrow to skin wound. Methods Porcine MSCs were isolated from bone marrow and grown in vitro. DAPI labeled MSCs were injected into bone marrow of the experiment animal. Full thickness wounds on the pig back were made by cutting. At 7,14 days after injury,tissues were harvested and observed under microscope. Results At 7,14 days after injury, DAPI-positive cells could be observed . The positive cells were more than that of control group at 7 days after injury. But at 14days after injury, DAPl-positive cells were decreased. The number of positive cells have no difference between 7 and 14days in control group. Conclusion Porcine MSCs may migrate from bone marrow to skin wound in vivo.
