目的:建立高效液相色谱法同时测定承德产山楂叶中牡荆素葡萄糖苷、牡荆素鼠李糖苷、金丝桃苷和槲皮素含量的方法。方法:色谱柱为Agilent ZORBAX SB-C18(4.6mm×250mm,5μm);流动相为0.5%甲酸水-乙腈-甲醇-四氢呋喃溶液,梯度洗脱;流速1.0ml/min;检测波长370nm;柱温30℃。结果:牡荆素葡萄糖苷、牡荆素鼠李糖苷、金丝桃苷和槲皮素的线性范围分别为9.38-300μg/ml、15.9-510μg/ml、4.51-144μg/ml、0.28-9.0μg/ml;平均加样回收率分别为100.2%(RSD=1.8%)、99.6%(RSD=1.5%)、99.4%(RSD=2.0%)、100.8%(RSD=1.5%)。结论:含量测定结果显示不同产地山楂叶中四种黄酮类成分含量的差异较大(RSD为9.98-31.39%)。
Objective: To establish simultaneous determination of glucosyl-vitexin, vitexin-2″-O-rhamnoside, quercetin and hyperoside contents in Chengde Hawthorn Leaf by HPLC. Methods: The separation was carried out in Agilent ZORBAX SB-C18 column(4.6mm×250mm, 5μm) with 0.5% formic, acid methanol, acetonitrile and tetrahydrofuran as mobile phase in a gradient elution model( flow rate:1.0ml/min, detection wave: 370 nm, column temperature:30℃). Results: The linear range of glucosyl-vitexin, vitexin-2″-O-rhamnosid, hyperin and quercetin were respectively 9.38-310μg/ml, 15.9-510μg/ml, 4.51-144 μg/ml and 0.28-9.0μg/ml. The average recovery rate were respectively 100.2%(RSD=1.8%), 99.6%(RSD=1.5%), 99.4%(RSD=2.0%) and 100.8%(RSD =1.5%). Conclusions: The contents of 4 flavonoids in Hawthorn Leaf from different production place are quiet different(RSD: 9.98-31.39%).
