目的:研究3’-大豆苷元磺酸钠对大鼠脑缺血/再灌注损伤的保护作用,并初步探讨其保护机制与抗氧化作用的关系。方法:大鼠大脑中动脉栓塞,制作局灶性脑缺血/再灌注损伤模型,缺血后10min,于舌下静脉给不同剂量的3’-大豆苷元磺酸钠。再灌注24h后,腹腔静脉取血并分离血清,断头取脑,一组前脑做。TTC染色,测定脑梗死体积,另一组左脑制成10%的脑组织匀浆。比色法测血清及脑组织匀浆SOD、LDH的活性及MDA的含量。结果:与模型组相比,各刘量的3’-大豆苷元磺酸钠治疗组脑梗死体积减小;血清及脑组织SOD的活性显著升高、MDA含量均降低;血清LDH活性明显降低,组织LDH活性降低。结论:3’-大豆苷元磺酸钠对大鼠脑缺血/再灌注损伤具有一定的保护作用,其保护机制与3’-大豆苷元磺酸钠提高抗氧化作用、增强氧自由基的清除有关。
Objective :To explore the therapeutic effects of 3' -daidzein sulfonate sodium on cerebral ischemia/repcrfusion injury and the relationship between its mechanism and antioxidation. Methods:A rat cerebral isehemia/reperfusion injury was made through embolization of the middle cerebral artery, and the injury was treated with 3' - daidzein sulfonate sodium,using sublingual vein injection. The cerebral tissue was stained using the TI'C method,and the volume of infarction was investigated. Moreover,the activities of SOD and LDH,the levels of MDA in serum or tissue were measured. Results:After treatment, a reduction in the volume of infarction and an elevation in the activities of SOD in serum and tissue were observed. The decreases in activity of LDH in serum and MDA levels in serum or tissue were also observed. Conclusion:3' -daidzein sulfonate sodium has certain therapeutic effects on cerebral isehemia/repeffusion injury because it enhances the antioxidation and the ability of oxygen free radical clearance.
