目的:探讨不同浓度的TCDD对体外培养软骨细胞程序性死亡过程的影响。方法:胰酶消化法体外培养兔的软骨细胞;软骨细胞株暴露于二英(TCDD)24 h(浓度为1×10^-10-1×10^-8mol/L);光镜和透射电镜观察软骨细胞株细胞形态变化;流式细胞仪检测软骨细胞株细胞凋亡率。结果:1×10^-10-1×10^-8mol/L的TCDD诱导软骨细胞出现核固缩、碎裂和凋亡小体,并随TCDD浓度的增加而更明显;TCDD诱导的软骨细胞凋亡呈剂量依赖效应,1×10^-10-1×10^-8mol/L浓度的TCDD作用24 h后,软骨细胞株凋亡率分别为11.75%±2.41%,19.5%±2.75%,27.5%±3.45%,对照组为5.21%±2.46%,差异有显著性(P〈0.05)。结论:TCDD可以诱导体外培养的软骨细胞发生细胞凋亡。
Objective: To investigate the effect of 2, 3, 7, 8-tetrachorodibenzo-p-dioxin (TCDD) on the cell apoptosis in rabbit chondrocyte. Methods: The chondrocyte from rabbit knee were cultured with TCDD (1×10^-10-1×10^-8mol/L) for 24 hours. The light microscope and transmission electron microscope were used to measure the appearance of chondrocyte. The apoptesis in chondrocyte was detected with flow cytometry. Results: TCDD (1×10^-10-1×10^-8mol/L) induced cell nucleus to dry up and shrink, broken to pieces and appearance of apoptotic body. The rate of apoptosis in chondrocyte induced with TCDD ( 1×10^-10-1×10^-8mol/L) were 11.75 % ± 2.41%, 19.5% ± 2.75%, 27.5% ± 3.45%, compared with 5.21% ± 2.46% in control group ( P 〈0.05) .Conclusion: The apoptosis may be up-regulated by TCDD in chondrocyte.
