目的研究富硒麦芽粉水提取物是否具有抗人胚肺成纤维细胞衰老的作用。方法过氧化氢处理体外培养的人胚肺成纤维细胞1h,然后加水提取物或预先处理,然后进行检测。CCK8法检测细胞增殖,流式细胞仪检测细胞周期及活性氧自由基,衰老相关的β-半乳糖甘酶染色检测细胞衰老,免疫印迹法检测细胞衰老相关的蛋白表达。结果本研究以过氧化氢处理作为细胞衰老的模型,使用富硒麦芽粉水提取物对细胞增殖没有影响的浓度处理后,可明显促进其增殖。进一步研究发现,其作用是降低细胞内活性氧自由基水平,减少衰老阳性细胞,减轻过氧化氢引起的G。期阻滞作用,降低衰老相关蛋白p21的表达。结论富硒麦芽粉水提取物具有较明显的抗人成纤维细胞衰老的作用。
Objective To explore the possible antiaging effect of water extracts from selenium-enriched malt powder (WSM) on human lung embryonic fibroblast cells. Methods Human lung embryonic fibroblast cells cultured in vitro were treated with hydrogen peroxide for 1 hour before incubation with WSM or pretreatment; CCK8 was applied in detecting the cell proliferation, flow cytometry in detecting cell cycle distribution and accumulation of intracellular reactive oxygen species (ROS), senescence-associated beta-galactosidase staining in detecting cellular senescence and immune-blotting in detecting protein expression. Results The model of cellular senescence induced by hydrogen peroxide showed out an obvious prolif- eration after the treatment with WSM at non-growth-inhibitory concentration, further research revealed that the mechanism lay in its effect in reducing the ROS level, decreasing the number of senescent cells, mitigating retardation at G1 phase in- duced by hydrogen peroxide and downgrading the aging-related p21 expression. Conclusions WSM is of obvious antiaging effect on human fibroblast cells.
