目的探求脑胶质瘤RUNX3的表达及生物学意义。方法手术切除脑胶质瘤标本55例相对制成石蜡组织微阵列,运用免疫组织化学方法检测RUNX3在上述55例癌灶和其中19例癌旁相对正常脑组织中的表达情况和分布特点,并分析其表达与患者临床病理特征之间的关系。结果胶质瘤中RUNX3蛋白表达率为38.2%(21/55)较相对正常脑组织中表达率94.7%(18/19)明显下降,差异有显著性意义(P〈0.05)。RUNX3蛋白表达无性别、年龄及病理分级间差异,但在病灶直径≥3 cm及生存期〈3年的胶质瘤患者中,RUNX3蛋白表达显著低于病灶直径〈3 cm及生存期〉3年的患者(P〈0.05)。RUNX3在不同类型脑胶质瘤中呈现差异表达。结论RUNX3蛋白表达下调可能与胶质瘤进展及患者的预后有关。
Objective To explore the expression characteristics and biological significance of RUNX3 in different glioma.Methods Surgically removed 19 relatively normal brain tissues and 55 glioma tissues were collected and embeded with paraffin to make tissue microarray using high- throughput tissue chip technology. Expression of RUNX3 in these tissues was detected by immunohistochemistry and the relationship between RUNX3 expression and clinical pathological features of glomia patients were analyzed. Results RUNX3 protein was highly expressed in relatively normal brain tissues( 94. 7%) compared to glioma tissues( 38. 2%,P〈0. 05). The RUNX3 expression was not obviously related to gender,age and pathologic stage,but was statistically corelated with tumor size( diameter≥ 3 cm) and patients' survival( overall survival 3years)( P〈0. 05) and the expression of RUNX3 was different in different types of glioma. Conclusion RUNX3 gene expression is related to the tumor size and survival,which indicates important molecular events in glioma progression.
