制备了一种新型纯化抗体的二乙氨乙基葡聚糖接枝离子交换介质,并研究了其对抗体的静态吸附及传质动力学。结果表明,二乙氨乙基葡聚糖接枝介质在低离子交换容量时对抗体有很高的吸附容量和传质速率,饱和吸附容量最高达到222mg/g湿介质,传质速率(De/D0〉0.44)是商品化未接枝介质DEAESepharoseFF的3倍(De/D0〈0.12),是分离纯化抗体性能良好的离子交换介质。
Sepharose FF was modified with diethylaminoethyl dextran (DEAE-dextran) to obtain a novel anion-exchanger for antibody separation and purification. Adsorption equilibria and kinetics of x-globulin on the DEAE-dextran grafted anion-exchangers with 4 different ionic capacities were investigated. It was found that both the adsorption capacity and uptake kinetics of x-globulin were significantly improved after DEAE-dextran grafting, even at low ionic capacities. The maximal saturated adsorption capacity of x-globulin reached 222mg/g wet resin. Especially, the uptake rate (De/D0〉0.44) was three times greater than commercially available non-grafting DEAE Sepharose FF resins (De/D0〈0.12). The research thus proves DEAE-dextran grafted Sepharose is an excellent anion exchanger for antibody separation and purification.
