目的探讨中药单体黄芩苷对全脑缺血/再灌注(I/R)大鼠海马神经干细胞(NSC)增殖和认知功能的影响及其作用机制。方法将45只SD大鼠,随机分为假手术组(S组)、全脑I/R组、全脑I/R+黄芩苷组(I/RB组),每组15只。全脑I/R模型采用双侧颈总动脉夹闭合并低血压。I/RB组在造模后2 h腹腔注射黄芩苷(200 mg/kg,1次/d),S组及I/R组在相同时间点给予相同体积的生理盐水腹腔注射。造模后14 d采用Morris水迷宫观察大鼠空间学习记忆能力,免疫组织化学法观察海马Brdu表达,Western印迹检测海马脑源性生长因子(BDNF)的表达。结果与I/R组相比,I/RB组改善了全脑缺血大鼠空间学习记忆能力的损伤(P<0.05),增加了海马齿状回(DG)Brdu阳性细胞的表达数目(P<0.01),同时明显上调了海马BDNF蛋白的表达水平(P<0.01)。结论黄芩苷能促进全脑缺血大鼠海马NSC的增殖和认知功能的改善,机制可能与上调全脑缺血后海马BDNF蛋白的表达有关。
Objective To investigate the effect of baicalin on proliferation of hippocampal neural stem cells and cognitive function in global cerebral ischemia/reperfusion(I/R) rats, and its mechanisms.Methods A total of 45 SD rats were randomly divided into sham oper-ation(S group), global cerebral I/R,global cerebral I/R+baicalin treatment (I /RB) groups.The I/R rat model was induced via the bilat-eral occlusion of the common carotid arteries and hemorrhagic hypotension .The proliferating neural stem cells were labeled by Brdu .Morris water maze test was executed to detect spatial learning and memory .Immunohistochemistry of Brdu was used to detect the proliferation neural stem cell.Western blot was implemented to detect the expression of hippocampus BDNF .Results Compared to I/R group ,baicalin im-proved spatial learning and memory damage (P<0.05),increased the number of proliferous neural stem cells (P<0.01) and the expression of hippocampal BDNF in I/RB group rats(P<0.01).Conclusions Baicalin could improve the proliferation of hippocampal neural stem cells and ameliorate the cognitive function of the rats with global cerebral ischemia /reperfusion , which might be associated with its promotion on the hippocampal BDNF expression .
