以羽衣甘蓝(Brassica oleracea var.acephala)SmbSmb自交不亲和系为试验材料,分离了控制花粉自交不亲和基因——S13-b单倍型富含半胱氨酸基因(SCR13-b),GenBank收录号为EF577028。SCR13-bcDNA序列含有237bp的开放读码框(ORF),编码一个含78个氨基酸的蛋白。SCR13-b sDNA含有一个758bp的内含子,内含子5’供体位点和3’受体位点边界序列符合GU-AG规则;DNAblot分析表明SCR13-b在基因组中只有单一拷贝;授粉生物活性分析试验显示原核表达SCR13-b融合蛋白能够启动SI反应,抑制杂交亲和花粉的萌发和生长。
The S13-b-IOcHS cysteine-rich gene (SCR13_b) was isolated from pollen of ornamental kale (Brassica oleracea var. acephala) S13-b homozygotes, GenBank accession number is EF577028. The SCR13-b cDNA sequence had an ORF of 237 bp, encoding a 78-aa protein. The genomic SCR13_b sequence contained a 758 bp intron with boundary sequences of GT at 5' site and AG at 3' site, which complied with the typical ‘ GT-AG rule’ in plants. Genomic DNA blot analysis showed that SCR13-b was a single copy gene in S13-bS13-b lines. Pollination bioassay showed that prokaryotic expressed recombinant SCR13-b can induce SI response and inhibit cross pollen germination and growth.
