目的探讨抑制Nogo蛋白受体(NGR)对糖尿病(DM)大鼠视网膜神经节细胞(RGC)的保护作用。方法尾静脉注射1%链脲佐菌素50mg/kg建立30只SD大鼠DM模型,分别向双侧眼球玻璃体腔内注射NGR小干扰RNA(siRNA)病毒10μl(A组,10只)、病毒稀释液10μl(B组,10只)和病毒阴性对照液10μl(C组,10只);另取10只正常SD大鼠(D组),同法注射病毒稀释液10μl。12周后,免疫组化法检测NGR在视网膜中的定位,Western blot检测NGR的表达,DNAladder检测各组细胞凋亡情况。结果 NGR主要表达于RGC。与D组相比,B、C组NGR表达明显上调(P0.01),A组表达无明显变化(P0.05)。B、C组RGC呈现明显的DNA ladder带,而A、D组未见明显DNA ladder带。结论抑制NGR的表达可减少DM的RGC凋亡。
Objective To explore the protection of inhibiting NGR on retinal ganglion cells (RGC) in diabetic rats. Methods The rat model with diabetes mellitus(DM) was established by intravenous injection of streptozotocin 50 mg/kg in 30 SD rats, which were equally divided into 3 groups of A(injecting NGR siRNA virus 10/μl into the vitreous cavity of both eyes), B(injecting diluted virus 10μl),C(injecting virus-negative contrl solution 10μl). Ten normal SD rats(group D) were injected with diluted virus 10μl in the same manner as in DM rats. Twelve weeks after injection, the localization of NGR in the retina, NGR expression and RGC apoptosis were detected by immunohistochemistry, Western blot and DNA ladder, respectively. Results NGR mainly expressed in RGC. Compared with group D, NGR expression was up-regulated in groups of B and C(P〈0. 01), which in group A was not significantly changed. DNA ladder was shown in RGC in groups of B and C, which was not seen in groups of A and D. Conclusion Inhibition of NGR can reduce RGC apoptosis in DM rats.
