目的:探讨切割穹窿海马伞侧大鼠海马与正常侧海马中PEBP mRNA的表达差异。方法:取6只大鼠,于切割右侧穹窿海马伞后第7d,应用寡核苷酸探针原位杂交技术观察PEBP mRNA在切割侧和正常侧海马中的表达差异。结果:切割侧锥体细胞层和齿状回颗粒层PEBP mRNA阳性细胞数量与正常侧比较无明显差异(P〉0.05),但切割侧的杂交信号强于正常侧(P〈0.05);而在齿状回门区和颗粒下层,切割侧PEBP mRNA阳性细胞数量多于正常侧,杂交信号也强于正常侧(P〈0.05)。结论:结合本课题组以往的工作,切割穹窿海马伞后海马内PEBP mRNA的表达上调,可能与海马内神经干细胞向胆碱能神经元的分化有关。
Objective:To investigate the difference of PEBP mRNA expression between the fimbria-fornix transected and normal side hippocampi.Methods:Six SD rats were selected and on the 7th day after fimbria-fornix transected,the method of In Situ Hybridization(ISH) was used to detect the difference of PEBP mRNA expression between the fimbria-fornix transected and normal side hippocampi.Results:In the area of pyramidal layer and granular layer of dentate gyrus,the positive cell number in the transected side and normal side showed no significant difference(P0.05),but the hybridization signal in transected side was higher than the normal side(P0.05).In the area of hilus and subgranular layer of dentate gyrus,the transected side's positive cell number was much than the normal side's,and the hybridization signal in transected side was also higher than the normal side(P0.05).Conclutions:Combined with our previous studies,the up-regulated PEBP mRNA in the fimbria-fornix transected side hippocampus maybe associate with the neural stem cells differentiating into cholinergic neurons in hippocampus.
