目的:探讨缺失突变型IκBα对HL-60细胞凋亡和分化的影响。方法:通过脂质体技术,将pCLX-IκBα△N重组载体质粒DNA转移到HL-60细胞并于转染48小时和72小时后,分别分析AnnexinⅤ和PI以及CD11b的表达。结果:转染后,AnnexinⅤ+/PI-细胞数增加。其中,转染72小时后的AnnexinⅤ+/PI-细胞数高于转染后48小时的AnnexinⅤ+/PI-细胞数。然而,无论是转染后48小时还是72小时,表达CD11b的阳性细胞数与未转染细胞的CD11b阳性数几乎无差别。结论:短暂表达IκBα△N cDNA能够诱导HL-60细胞发生凋亡,但似乎并不影响该细胞向粒系的分化。
Objective :To explore the effects of mutant IκBαt on the apoptosis and differentiation of HL -60 cells. Methods: The plasmid DNA of recombinant pCLX -IKBctA N vector was transferred into HL- 60 cells mediated by liposome followed by analyzing the expression of Annexin V, PI and CD1 lb after both transfection 48 and 72 hours. Results: The number of Annexin V^+PI^- cells was increased after transfection, which was higher in 72 hour of transfection than that in 48 hour. However, the positive cells of CD1 lb expression were almost no difference between transfected and untransfected HL -60 cells no matter when it was in 48 or 72 hour after transfection. Conclusion: Brief expression of IκBαΔ N eDNA can induce apoptosis of HL - 60 cell, but it almost can not affect the cell differentiation.
