探讨羟基磷灰石(HAP)纳米粒子负载阿霉素后的体外抗肿瘤活性。通过纳米粒子与人肝癌Bel-7402细胞和人正常肝细胞L02的体外培养,研究阿霉素负载HAP纳米粒子的心肌毒性,以及对肿瘤细胞凋亡的影响。MIT(四甲基偶氮唑蓝盐)法检测表明,阿霉素负载HAP纳米粒子对Bel-7402细胞的抑制率为95.22%,明显高于HAP纳米粒子(16.26%)和阿霉素(83.36%)(P〈0.05)。倒置相差显微镜观察显示,阿霉素负载HAP纳米粒子比单用阿霉素有更大的肿瘤抑制效应,并降低心肌毒性。
The aim of this paper was to study the anti-tumor activity in vitro of adriamycin-loaded hydroxyapatite (HAP) nanoparticles. The cardiac toxicity of adriamycin-loaded HAP nanoparticles was studied by the in vitro cell cultures. MTT ( Blue methyl thiazolyl tetrazolium salt) assay showed that the inhibition rate of adriamycin-loaded HAP nanoparticles was 95.22%, which was significantly higher than HAP nanoparticles (16.26%) and adriamycin (83.36%) ( P 〈 0.05). Inverted microscope observations also showed that the inhibition effect of adriamycin-loaded HAP nanoparticles on cancer cells was better than that of anti-tumor and the cardiac toxicity of adriamycin was reduced as well.
