目的 探讨基质衍生细胞因子-1α( SDF-1α)在根管预备后的根管管腔环境下诱导内源性细胞归巢的能力.方法 临床收集因正畸需要拔除的前磨牙,制备成长5 mm的根管段共30个,分为实验组及对照组各15个,实验组根管段内植入复合100 ng/ml SDF-1α的多肽水凝胶胶原支架,对照组根管段内仅植入多肽水凝胶胶原支架. 选取15只裸鼠,于左右两侧皮下组织分别植入对照组、实验组根管段样本,4周后取材,对样本进行组织切片观察. 结果 实验组根管段内见大量细胞和血管的结缔组织形成,对照组根管段内见均质状疏松组织,未见细胞、血管等新生组织. 结论 SDF-1α在根管预备后的根管管腔环境下具备诱导内源性细胞归巢的能力。
Objective To explore the capacity of stromal cell derived factor-1α( SDF-1α) inducing endogenous cells homing into the prepared root canals.Methods Extracted preomolars for orthodontic treatment were cut into 30 roots segments with a length of 5 mm. Thirty root canals were divided into experimental group(n=15) and control group(n=15).The root canals of experimental group were filled with 100 ng/ml SDF-1αcombined with peptide hydrogel scaffold,and of control group were filled with peptide hydrogel scaffold only. Fifteen nude mice were selected and implanted subcutaneously on the left side and right side with the root canals in the experimental group and control group respectively.The samples were taken out 4 weeks later,and then made as tissue slices for observation.Results A large number of substancial cells and blood vessels were observed in the root canals in the experimental group.In the control group,there was homogeneous open texture found in the root canals, but no neogenetic cells or blood vessels were observed.Conclusion SDF-1αmay induce endogenous cells homing into the prepared root canals.
